Silver staining is used to detect proteins after electrophoretic separation on polyacrylamide gels. Among the various protein detection methods, the technique of silver staining has gained wide popularity because of its higher sensitivity (in the low ng range) than that of Coomassie blue. Meanwhile, it can be achieved with simple and inexpensive laboratory reagents and no complicated and expensive hardware for the readout is required.
Silver staining now is commonly used in the field of environmental science, botany, breeding science, etc., for example, it aids the visualization of electrophoretically separated proteins in Histological characterization research. It can be also used for diagnostic microbiology, karyotype analysis, genomic and proteomic analysis and many other applications due to its high sensitivity and low cost.
During the procedure of silver staining, many factors may have influence on the final staining results. Has it ever occurred to you that the water you use for the overall experiment has played a very important role in deciding whether it is a successful staining or not?
For example - for optimal sliver staining results, the level of chloride ion (Cl-) in the water used must be kept as low as possible. If the amount of chloride ion present in the water or reagents reaches 2 ppm under normal conditions, precipitation is likely to take place and a big haze appears on the gel surface at the development stage. A terrible background results in a failure of your experiment.
One thing you must know- silver staining is very sensitive to trace impurities in water. It is critical that water with a resistivity greater than 15MΩ/cm must be used.
In brief, only EDI pure water or ultrapure water can be used in silver staining, while RO pure water cannot give you a satisfactory result!
